九一八事变后，抗日呼声愈发高涨，苏海龙和黄显声在沈阳城打响抗击日寇的第一枪。养父与亲人相继离世，苏海龙走出悲痛，积极组建抗日队伍，随后和邓铁梅一同加入东北抗日革命军。在张婉婷的影响与帮助下，苏海龙光荣加入中国共产党，带领部队沉重打击日本关东军。日本关东军多次围剿苏海龙所在部队。随着抗日斗争形势渐趋严峻，东北抗联奉命陆续前往苏联远东地区休整。苏海龙奉命前往延安抗大学习，学成后被派往苏联远东第88步兵旅。他多次率小分队潜入东北，对日本关东军开展渗透和侦查工作，成功炸毁虎头要塞那门号称“亚洲第一炮”的巨型榴弹炮，为后续总攻扫除了障碍。
